Are you active or reserve? Generally speaking, you will qualify yearly on the M-16. You may get a chance to shoot the M-9 (9mm pistol) as well, but that is generally for Chiefs, officers, and CSE.
During miltary training year, you will most likely be put in a class for crew served weapons. Either teh .50 cal, 240B, or MK-19. You will go through a classroom environment, then go the range and shoot.
Depending upon your unit, you may get a chance to shoot the AT-4 (freaking sweet) and the M-203(grenade launcher)
Yes, it is mandatory, a Seabee who can't fight is pretty worthless to the unit.

ps. At FEX, you will be on either the crew served weapon you had trained on or being going on patrol, and you will use blanks.

Or if you are really good you can work in the comm section.

Then you can reach to the DASC and call down the thunder on your enemies.

With all due respect there is two things that every good Seabee must remember about communicators.

1. Comm, You can talk about us but you can't talk without us.

2. If you are in the field and you are not communicating...then you are only camping.
